      K powder hair detection reagents use antibody-antigen specific binding response and immunofilm tomography technology to detect ketamine (KET) in human hair by immunocompetitive inhibition. That is, the protein-labeled ketamine vector competes with the ketamine in hair to compete with the antigen binding site on the limited antibody. The concentration of ketamine drugs added to the sample was different from T-line fluorescence, and the change in fluorescence values reflected the concentration of ketamine drugs.

    Six, the use of preparation

    Seven, hair extraction

      According to the "Toxic Person Hair Sample Testing Specification" to extract hair samples using the tooth shear from the subject's head (pillow) to collect the appropriate amount of hair (when no hair can choose sweat or armpits) sampleinto into a special sampling bag, seal hair and record sample information on the sample bag.

    Eight, hair pretreatment

      1. Check the sealing status of the hairs sent for inspection and register and check the sample information recorded on the sampling bag;

      2. Cut the collected sample into 1 mm -2 mm thin segments using scissors;

      3. Take the appropriate amount of cut hair samples placed on weighing paper, marked with a marker pen number;

      4. Take a 1000?L pipette to add 400?L hair treatment fluid A to the centrifuge tube with a sample, and then place the centrifuge tube in the hair front treatment instrument for 5 minutes;

      5. Remove after the reaction, take 1000?L pipette to add 400?L hair treatment b to the centrifuge tube;

      6. After oscillating mixed, spare after room temperature.

    Nine, sample testing

      1. Please read the instruction manual of dry immunofluorescent analyzer carefully before testing;

      2. Tear the aluminum foil bag open and remove the reagent plate, marked. With a matching sample straw to absorb the well-handled hair treatment fluid, vertical drop plus 2-3 drops of reagent plate plus sample hole, to react 5min after insertion into the "dry immunofluorescent analyzer" card tank, for detection;

      3. At the end of the test, the instrument screen displays the test results and automatically prints the inspection report and reference range.

    TwelveKetamineHair detection precautions

      1. Hair treatment fluid A and hair treatment liquid B is corrosive, please seal as soon as possible after use, wear gloves to avoid direct contact with the skin;

      2. Testing reagent board items, batch number identification items, batch numbers consistent, not mixed;

      3. When the pipette absorbs different liquids, it must replace the sampling nozzle to avoid cross contamination;

      4. Hair as far as possible cut, to avoid hair too long treatment results are not ideal;

      5. The amount of cut hair placed in the centrifuge tube must reach the 0.5 tick mark shown in the pipe wall label;

      6. If sample contamination is suspected, apply a new sample retest.
